Isolation and Potency Testing of Bacteria Producing Indole Acetic Acid (IAA) from The Rhizosphere of Bamboo (Bambusa Sp.)

Abstract
Bamboo (Bambusa sp.) is known as “giant grass” due to its rapid growth and high adaptability to various environmental conditions. The bamboo rhizosphere produces root exudates that create an ideal habitat for the growth of functional microorganisms. One of the roles of rhizobacteria is to act as a biostimulant that produces the hormone auxin. The ability of bamboo rhizosphere bacteria to produce Indole Acetic Acid (IAA) is still not well understood. This study aims to isolate and test the ability of bamboo rhizosphere bacteria to produce IAA. The research was conducted at the Soil Laboratory of the Faculty of Agriculture, University of Riau, Binawidya Campus, Pekanbaru, from October to December 2025. The study was conducted in two stages. The first stage involved the isolation of bacteria from the bamboo rhizosphere, purification, observation of colony morphology, and cell staining. The results showed eight isolates, all rod-shaped, comprising both Gram-positive and Gram-negative bacteria. The colonies were cream and milky white in color. Qualitatively and quantitatively, all eight isolates were capable of producing the IAA hormone. Isolate RBLP2 produced the highest level of IAA at 179.03 ppm.
